The use of molecular markers in the selection of wheat genotypes obtained from in vitro cultures
Summary.

Background. Leaf rust caused by Puccinia recondita f. sp. tritici is one of the most dangerous fungal diseases as it causes significant loss to common wheat (Triticum aestivum L.) yield. Breeding for resistance is the most effective method of controlling fungal diseases of cereals. Biotechnological methods such as molecular markers and the haploidisation technique shorten the breeding cycle and increase the efficiency of selection. The aim of the study was to identify the leaf rust resistance gene (Lr19) in wheat genotypes obtained in in vitro cultures.

Material and methods. The plant material consisted of haploid plants and spontaneously doubled haploids from generation F1 hybrids, which were obtained by crossing reference genotypes with the Lr19 gene (T36, T39) and well-yielding varieties ‘Ozon’ and ‘Hondia’. Xwmc221 and GB markers were used for identification of the Lr19 gene. 58 genotypes with the haploid number of chromosomes and 39 spontaneously doubled haploid plants were tested.

Results. The presence of Xwmc221 marker (200bp) was observed in 29 haploid plants and in 24 spontaneously doubled haploids. The presence of GB marker (130bp) was observed in 27 genotypes with the haploid number of chromosomes and in 27 spontaneously doubled haploid plants.

Conclusions. The analyses proved that selection based on molecular markers is possible at the haploid level, which enables the reduction of costs and length of the breeding cycle.
